Could be water on the knee. See a doctor and exercise your legs.

The pain is caused from a knee injury or may be you have arthritis. Or something along those lines. Keep moving and it wont hurt. When you are still try a warm wash cloth or heat to loosen up the muscles. Ibuprofen may also help with inflammation.

If you haven't injured it recently, it could be arthritis. Check with your doctor, usually the only way to know for sure is to have the knee x-ray'd. Before you get up after sitting for a while, bend and unbend the knee a few times to help loosen it up. If it's painful, ibuprofen and an ice pack will help.
